Training tips for 500km cycle ride! Please help!

Hi all!
I've signed myself up to do a 500k charity cycle ride through Vietnam and Cambodia in Nov 2012. I currently cycle to work 5 days a week (roughly 5 miles a day) and try to go on a decent ride most weekends (between 20-40 miles). I currently have a beautiful Pashley Tube Rider but I'll be getting a mountain bike on pay day as a road bike just isn't going to cut it!
I'm relatively fit but this is going to be a mammoth challenge so could do with any extra tips you lovely people may have!
All I want to do it to complete the 500km in 1 piece, still breathing and (this may be a long shot) still able to feel my backside!
Any hints and tips would be greatly appreciated!
