Most LBSs, in my experience, discount bikes little, if any, unless you happen on a sale, or a model-year-end clearance. Those around here that discount at all take $50-$100 off depending on the price point. I've tried for greater discounts than that, without success, but I'm not really much of a negotiator. Markup on bikes is not that great, or so I hear; most of a shop's profit comes from accessories and service.
