One addition to the above. After you loosen the lock nut on the valve stem, give it a quick tap as presta's tend to become sticky.

Air will go into the tube when the pressure in the pump exceeds the pressure in the tube, not before. Sometimes if the valve is a little sticky, pressure in the pump will get very high before the valve releases.
