Hey,
I just learned to lose one U.S. pound (.454 kg), you must burn 3,500 more calories than you take in as food. This chart below is for one hour of exercise, and for 210 lbs.

Now if just I could run 12 mph for one hour, I'd be set!
Actually, I'd like to be able to run one mile at that speed.
Calories burned per hour while running seems to vary depending upon where you read it. I've read conservative estimates such as 110 per mile. My treadmill tells me I burn about 150 calories per mile at ~6mph, 1% incline. If I bump up the incline to 10%, I get 200 calories per mile at a 6mph pace. I can maintain that pace for an hour, so if my treadmill is accurate (somewhat doubtful, since it doesn't even know what I weigh), I'm burning 1,200.
One of the workouts I do when I cannot get motivated to do anything else or have little time, is to get on the treadmill and do a 500 calorie workout. I try to vary the incline and speed to see how fast I can get to 500 cals. If I push it (running intervals of 8mph at 10% incline), I get there pretty fast. And it's pretty tough for a 2.5 mile workout.
