I have been doing well in the mountain bike division of our local triathlon series but I would like to enter in the regular division. I have been riding close to 20MPH on my Trek 4300 during the race. If i switch to an aluminum road bike how much speed improvement can I expect?

Hard to say, but if you're running fat tires, I would guess at least a few(3 or so) mph by switching.

Depends on both bikes:
1) MTB with knobby tires, and no aerobars to a road bike with racing tires and clip on aerobars, the difference could easily be 3 mph.

2) MTB with high pressure slicks, dialed in aero postion with clips on, and a road bike without aerobars, the road bike could even be slower.

IS quad pain above the knee normal when switching to a tri bike from a road bike or is my setup wrong

I don't know but 20 mph on a mountain bike sounds pretty damn good to me.

Sounds like the wrong setup to me... Check saddle height to make sure that you can extend your leg (but not over extend it! it should have a slight bend when you are at the bottom of your stroke). Also check cleat position (should be centered over the ball of your foot)
