I cycle 4-5 times a week and try to push myself pretty hard most times. My legs hardly every get sore and if they do it is very little. I ran a few short sprints the other afternoon and the next moring on my ride my legs could really feel it and by that afternoon they were fairly sore. Is the because of the use of more fast twitch muscle fibers and do any of you run and ride both?

Oh yeah. I have now started training for triathlons next summer and the first week that I did some jogging and running I was sooo sore. Give it a week and it'll get better.

I just started running three days a week, doing the coopers test (1.5m in 12 mins). My muscles are so sore right now, I literally cant go up/down a flight of stairs.

Delayed onset muscle sorness is believed to come mostly from eccentric muscle contraction which is the phase where the muscle is lengthing(stretching) under stress (load). In road biking there really isn't any eccentric loading, which is why when you ran which does involve this phase to a greater extent you got sore, generally your body will adapt to the stimuli that caused the soreness within a couple workouts. The best thing you can do for this is to go for a recovery ride the next day say 40-60 minutes at 65% HRM.
