I think my butt is simply a slow learner. It takes me several 75 mile rides (three or four usually does it) with a new saddle to determine if it's comfortable for long distance rides of a century or more. I don't start out riding that distance with a new saddle though. I work my way up to it in 20 mile increments. I'll do 10 miles for the first ride then bump it to two rides at around 30 and then a couple of more rides at around 60 and then go to around 75. Some saddles take to me (or vice versa) and others are so excrutiating after the first few rides that I figure it's just a bad fit. I had that experience with a Specialized BG saddle. Lately I've been having good luck with Selle Italia Flite-shaped saddles and have been going back to them so I haven't tried other new saddles lately.
