So, I am getting ready to finally glue mine on tonight, and remembered that tread patterns are often oriented differently. What is the correct way for Grifo Challenge tires to be glued on? Like the diagram below?

it doesn't matter. Bicycles don't make enough power to really challenge tread design.

I have my Tufo with the chevron looking forward as in your Front tire, both of them.
Look at some of the pro's pictures.
