I use a Deore LX 9speed 11-32 on my "road" wheel with my Ultegra STI shifters. Of course, I have to use a Deore XT (or other mountain) derailer to be able to handle the 32t cog.
Yes. Just ratios are different. You just have to watch der capacity.

The ATB cassette will have larger jumps between cogs affecting cadence more severely when shifting. Instead of 1-2 tooth changes you will see up to 4 tooth changes.
I tuned in an ultegra short cage derailleur with a 11-32 cassette on my road bike a while back for a big hill climb.
I had to shorten the chain a bit for it to work which made it where I had to drop off the 53 before the 28 tooth cog in the rear but by then your are cross chaining anyway so you should be off the 53.
It will take a bit of know how to tune it and get it right but it can be done.
