I have an old Falcon Ernie Clements which is currently running on 27" Mavic rims. Is it possible to replace these old type wheels to modern 700 size? If so, what are the problems I need to overcome?

Brake calipers. Maybe you can position them 4mm closer to the hubs, but if not, you might have to change them out also.

In addition to brake reach, if you're switching to a more modern rear hub, you'll need to increase the spread of the rear triangle.
Fwiw, I first tried 700c wheels on my 60s Falcon San Remo, but the thing I found most annoying was that the huge amount of space between the front fender and the front wheel. I could adjust the rear with a spacer, but the front wasn't going to work. I put a set of 27" wheels on instead, and it's doing fine.

Over the years I have freely swapped 700C wheelsets into framesets designed for 27", with good results. If you have an extra 4mm of brake slot length, you can make the change.

Dremel tool if the brake caliper slots can stand it, but be very careful.
Or
Buy long reach calipers, but you still may have to drill out if there's a mounting difference (recessed vs long bolt).
I'm successful 100% of the time switching, but sometimes that means doing nothing but adjusting the brake shoes, and other times, it's different calipers and some metal work to be done.

+1 I've laid off the switch for my SS builds, unless they absolutely have to have a deep v wheelset, and those that do generally have a color in mind. An alloy set of 27's is a nice looking option, and a whole lot easier.

I'm only thinking of changing because of the greater choice of tyre styles available for 700c wheels.

Mike, I just put a set of 700c wheels on my new-to-me Moto while waiting for a proper pair of 27x1" tires to arrive. Lets me ride the bike while I re-pack the wheel bearings and replace the 6 cog 13-26 Shimano 600 freewheel with a 7 cog 11-28 (anyone have a Regina/Shimano 600 freewheel tool I can borrow for a few minutes?:D)
