I've talked to a local shop about building a set of OP's on Ultegra hubs with an ultegra cassette. He says that he is an 'expert' wheel builder, and the price seems fair (rims: $60 ea, Hubs:120, Spokes: $25, cassette: $80, labor $45).
My questions are these: Is there a sort of qualification to be an 'expert' wheel builder? and does the price seem right?

The best way to judge the quality of a builders wheels is to talk to several people that have ridden his wheels for awhile. You can also get an idea by how many wheels they have built(Altho one could build a lot of crappy wheels you get better with time and a better builder tends to build more wheels)
If the labor is for both wheels, that is cheap, but then I would question ability as skilled labor cost more. $25 is about right for a set (32) of double butted spokes and a little high for straight guage. I would build it DB myself, nicer ride and a little lighter. Rims about right, we have them at $65. Cassette maybe a little high, we charge $70 for Ultegra cassette.

It would cost a total of $282 from Colorado Cyclist, delivered. No sales tax. Bought a set from them and put 8,000 miles on them.

My local store doesn't charge anything to build the wheel if I buy all of the parts from them.The guy builds nice wheels,maybe he just gives me a deal 'cause I've been going there for over ten years.
