I've been on five BNY5B rides (registered) and have seen many people riding without vests. If anyone minded I've never seen them show it. The only checkpoints seem to be the rest stops and a couple of the bridges. On the approach to the Queensboro Bridge they separate the non-vest riders and make them take another route across the river.

No race. No competition. Just have fun.
(I have a personal target of being over the Verrazano by noon, but that is so I can get back to Manhattan to ride some more.)
You can bail out at any time and take a subway back. Or wait for the SAG wagon. Or cut a few miles by skipping Astoria. 42 miles is not that bad a distance. The route is mostly flat. Biggest climb is the Verrazano. And there are plenty of stops.
You still might want to practice a bit first.

Looks like a great ride. There's a mailing list that will alert you when registration opens; I signed up.

Your average time per mile x 42 + 2 hours of walking your bike through mob bottlenecks.

There is some police support at major intersections.
Other than that, the only "protection" you have is that you are riding in large groups.
If you're not comfortable riding in traffic,
you may want to get some "city" miles under your belt first.
