Position. A comfort bike has more upright position and a road bike has more of an aero position.

That all depends on what you want to do with the bike. On rides over 10 miles or so, a true blue road bike is much more comfortable than any of the so called "comfort" bikes I have ever been on. Comfort bikes where designed to sell a bike to folks who just arn't sure they want a road bike. Then they buy one, ride it a while, then buy a road bike, or stop riding all togather!

If you wan't to ride miles, go ahead and get a proper fit on a road bike and be done with it. ;)

Many modern road racing bikes are designed for more extreme kinds of riding, eg highly aerodynamic positions. Check out the drop from the saddle to the bars.
Experienced riders can cope with 3-6" drop, but beginers, older folks, and those who prefer long touring rides prefer a more relaxed position.
The word "comfort" is a bit odd. The lower Trek design is what I would call a light or sport-touring bike. Its a road bike, but not for racing. Light touring bikes are high performance, they are just designed to perform differently to racing bikes.
There is another kind of comfort bike, which is basically a 26" wheel utility bike with suspension forks and seatpost. It is a pretty low-performance design.
