One of the spokes on my rear wheel brushes the derailleur when the chain is on the largest cog. What's the best way to go about fixing this?
Sounds like the derailleur cage is bent. If so, you'll need to either straighten it or buy a new derailleur.
Or your limit screws aren't set right, but its a 99% chance that your hanger is bent. Best to have a shop realign it as there is a special tool for this.

Well, it only touches two of the spokes. The other fourteen laced on its side glide by silently, unaffected. I can't tell by eyeballing it whether the hanger is bent. I guess off to the shop I go.

Don't panic! Last time this happened to me, the LBS charged me all of $15. Didn't take long, sure, but a bargain at twice the price, knowing it was done right.

Last time that happened to me it was the derailer hanger....Cheap compared to the derailer.
