I am thinking about trying to lace a single speed White Industries Standard ENO hub 32h to a 50mm carbon 24h tubular rim. Seems possible:

Would anyone advise against this for cross? Any suggestions?

If its a rim you won't otherwise use or care about, then I guess you could try. But only if you're an expert wheel builder as carbon low spoke count wheels require a very high spoke tension, which may or may not be tolerated by the hub flanges designed for lower tension, and a high level of building skill. It's not like building 3-cross metal wheels. And failure would really suck- not worth the negligible "benefit"- especially since the resulting substandard wheel probably wouldn't ride well anyway. If you're good enough to confidently do this, you probably wouldn't be asking the question--

shouldn't the number of holes match? 32h hub 32h rim? 24h rim 24h hub?

Yeah I probably do not have the skills to do it so would have it done professionaly.

Yes you are technically supposed to have the hole count match but there are ways around it.
