I just recently bought my first road bike (cannondale RW400) and love it, except for the seat...the pressure around my "special friend" is annoying while i'm riding, but it doesn't hurt or cause numbness. Keep in mind i've only logged around 4 hours total on it, in a few different rides. I'm wondering how long it takes to adjust to/break in seats (mine is a Bontrager CRZ sport) and how long i should keep making minor adjustments before i get a new seat.

I don't feel that one breaks in a saddle even if it is a leather Brooks saddle. You break in your bottom. Even when you are completely used to a saddle you should stand up periodically to get your weight off of your bottom. Every fifteen minutes is non too often and maybe more frequently if you have been pulling a long slow hill. In time you will do that automatically without even thinking about it. You should of course check to make sure your saddle is properly positioned and pitched.

Stock bikes often come with inferior saddles. It is one of the ways a manufacturer hits a price point. Start with a good high end saddle from say Avocet, San Marco etc and one that fits your riding style.

Next have a qualified person fit the bike to you. Saddle too far forward and you will have hand and shoulder problems from too much weight on the bars. Too far back and you will have too much weight over the back wheel and on the saddle. Their is a fine balance between saddle height, fore/aft adjustment and bar height.

Also just plain time in the saddle helps.

I'm a little ahead of you, but not an expert by any means. I found myself creeping forward on my saddle for whatever reason. I moved my seat forward and that seemed to help. Also, I found my "sit bones" and try to position them squarely on the "pads" towards the back of the seat.

Tonight, I tilted my seat up a little based on some of the comments I read on other posts. I'll let you know if it worked!!!
