I'm thinking of cycling from Cambride to Long Point on the long weekend: about 110 kilometers. Is there anything I should know about doing this ride? Any roads that are good to take? Any roads I should avoid at all costs?

I've done Waterloo to Rock Point, fairly similair distances I think. But can't give much route specific details since they're different directions.
The only advice I have would be to check google maps satellite view to make sure you don't hit gravel roads, GPS's and Googles routes tend to not know the difference between small paved roads and gravel roads.

Sounds like a good ride. I just googled directions, and it looks like there are plenty of back roads in that area. My wife and I bike the rail trail from Cambridge/Paris/Brantford a couple times a year - its a nice leisurely ride that would get you part of the way there, but I'd guess that you're probably more interested in a road route.

We've also ridden along Lake Erie from Port Burwell through to Turkey Point and beyond, which is in the same general area. Great, quiet roads. Not sure about the long weekend though - you'll probably want to stay off the beaten path.

Hope this helps - have a great ride!
