Bought an new RaceFace 44,32,22 Crankset the other day.
Via the placement of spacers the chainline is adjustable.(49,50,51mm)
Since I ride in the large chainring the majority of the pedal time, I am wondering if
setting it at 49mm would be more optimal.
Any thoughts?

What sort of crankset is it? if it's an External BB one, you need to follow the manufactures specs for the spacers.
If you have a Race Face BB, it is printed on it, otherwise look at the instructions provided, or on the manufactures website.
It depends on which rear sprockets you use the most. If you're using it mostly with the outermost few in the rear, moving it outboard might make more sense. Think on terms of which of the three will yield the best chainline based on the gear combinations you actually spend the most time in, and set the chainline to favor those, while keeping it reasonable for the others.
In any case 1-2mm either way won't make a word of difference.
