Patch: tube inflated at all?

when applying the patch to the tube, should the tire be inflated at all?

No. You don't want leaking air affecting the seal of the patch. Don't worry about what shape the patch will take on when you put air in the tube, because it will all conform to the inside of the tire anyway.
