Lateral flex

Can a rear wheel be checked for lateral flex if it is off the bike? When trued and tensioned can lateral flex be checked/known before putting the wheel back on the bike and riding it? What are the spoke tension specifications for a Bontrager SSR rear wheel in order to be tuned properly? Thanks in advance.

You can look here for some Bontrager specs but I would email customer service at Bontrager or check with your local Trek dealer to be certain of the correct tension specs. I heard they were just like the Race series of wheels but I'm not sure.

Sure, all you have to do is to stick the wheel in something else and then give the rim a sideways pull.
Flex only occurs when there's a load, and for most people the only reasonable way of loading a wheel is to stick it in a frame and ride it.

Sure, as long as you have another fixture available to hold the wheel while you give the rim a sideways pull and measure the displacement.
