I have two road bikes with downtube shifters. I want put 10 speeds cassette. I have 9 speeds chain. The 10 speeds cassette is working with 9 speeds chain or i need to buy 10 speeds chain? I have one ultegra 9 speeds groupset 9 speeds chain and 9 speeds cassette is worth i take out from my one road bike is cannondale R300 aluminum frame. Maybe 9 speed chain will work with 10 speed. Very close in size.
Since you already have the parts, go try it for yourself, then you will know for sure.

I have just installed a shimano 10 speed cassette on my 1987 Ricardo elite with shimano 6 speed DT shifters from 1987. It's very smooth (friction) shifting without dramas. Have to say I was surprised how well it works. Won't need to buy any dura ace DT shifters after all!

Whew, a rather old post. But, I've done 9s with Campy frictions shifters, and an Ultegra RD.

I got chain rub with the 3/32 chain, so I had to go with a 9s chain. Otherwise, no issues. I think the 9s cassette is much easier to shift and find gears than the original 5s freewheel. I'd expect 10s, or perhaps even 11s would shift reasonably well too. Keep in mind the derailleur jockey wheels are designed with a little float which helps with imprecise shifting.

With the 9s, essentially anywhere I park the lever, I'm in a gear.
