Most go clockwise because it's more scenic. Not too much climbing (see above), but the elevation makes it a bit harder. I've had good luck starting and ending at State Line (southern part of the lake), but you could really do it from anywhere. (I have never done it solo, but I've done the organized bikethewest.com ride twice.)
My best advice is probably to do it very early on a Sunday morning otherwise the traffic will really take all the fun out of this ride. EDIT: Saw that you will be there for a while, so a weekday would probably be even better.
