DT Aerolites?
Sounds like a sweet build with the Interlock's, but not the Alex's.
Maybe some Mavic Open Pro's if you can push the budget a bit.

Spokes: DT's. However, the issue here is the perpetual one in cycling-weight savings (and here we're dealing with rotational weight) vs. reliability. I've never used Tune hubs and I gather they're available in different iterations. Ditto with Alex and Interlock rims. Should I stick with tradition (DT all the way) or go the Tune route?

They make some kick-ass rims.
Yes, use the Tune's
They have some of the tightest tolerances of any hubs made, and they will last forever.
