Bought a new wheelset and need help

So..

My Mark V used to be super silent until i got some new wheels. I bought Deep V's and Formulas (of course). Ever since I put them on my rear wheel makes this weird noise. I keep my chain always at around 1/2 to an 1" of play so its not too tight. To top it off, the other night my came off... Luckily I was near the subway. Someone please help me.

Also, my chain gets loose and tight when my crank is in different positions. Someone help!

Noob questions: My chainline looks straight but how do I know if it really is or not?

Your chain ring is not centered

Check out sheldonbrown.com - his articles on doing a fixed gear conversion include info on both how to center a chain ring, and he has a whole article on chainline.

It's not that the chainring isn't centered, but many chainrings aren't perfectly concentric, and will therefore have positions where the chain is tighter and looser.

There is also a possibility that your chainring or cog is bent out of shape. Also, your chain could be stretched or not pulled to correct tension, generally it is a good idea to have a good amount of tension to avoid having your chain come off while riding.

you might be running too much chain tension.
you shouldn't have any 'tight' spots.

generally you should run your chain just tight enough so it doesn't fall off while riding.
