Do you mean using a backpack as your way of packing rather than using rack and panniers...YES. I did 2700 mile trip last year and 1700 mile trip this year with a backpack. Sleeping wherever my head landed for the night, generally town property, whether baseball dugouts, covered bridge over a rail trail, park pavillons, etc.

I will ride my bike a day or two to a trailhead, and go out for a multi-day hiking trip, if that's what you mean. I guess there are several interpretations of your question. I have a frameless backpack and a lightweight hiking style that makes this easy. I cycle in the same shoes I hike in, and use old-fashioned toe straps on the bike.

Do you transfer from panniers to backpack, or lash the pack to your bike with gear inside.... or? I would probably carry some stuff on the bike that I may or may not take out on the trail.

I transfer from panniers to backpack. I tried to make it work with the full pack lashed to the top of the rack, but I didn't like it--convenience and stability mainly. My pack is a 9 oz Gossamer Gear pack, made of silnylon, and it packs easily when empty.
