Looking to get a 2nd wheel for my trainer

I'm sure it doesn't need to be an exact duplicate of the regular wheel. Does it need to have the same cassette? Will having a different cassette cause the derailleur settings to be off?

nope. Well, not necessarily - as long as the cassette is close - so that you don't have to shorten/lengthen your chain.

I run a 12-25 on my road wheels, but a 13-26 on my cheapo Alex trainer rim. I don't have to adjust anything when I switch. Both cassettes run perfectly smooth.

+1 - exactly the same solution I use. I think as long as you have a cassette that is close - w/in 'a few' teeth on the top and bottom in order to have roughly the same chain tension - then you're good to go.
