No you don't have to take the chain off. I found it easier to with the chain on actually. They are so simple to use, I cut my chain cleaning time down quite I bit with that machine.

Its easier with the bike up right. If you don't have a bike stand, find a way to get the bike off the ground. I take it over to my dad's house, and tie the bike to the rafters of his carport. I guess that is a poor man's bike stand.

I just realized your link is for the cleaner only. I would suggest getting the combo, that way you have the brush as well. That brush gets in the spacings of the cassette and the chain better than a toothbrush.
