Bicycle is making weird sound when I ride it?!!

A few days ago my bicycle began to make this weird sound. The back wheel makes like a "TAKCH" sound about once every revolution. Surprisingly, when I don't sit on the bike (just pull it along while i'm walking), it does not make that sound; only when I sit on it. Apparently the weight is probably pushing down on something (I'm not heavy - 120lb). I checked the brakes and the chain, and they were as usual. I don't know what is causing that sound. Can someone please help me out.

Thanks.

P.S. My bicycle is a "Supercycle 1200" with 3 gears.
