Major shifting issues
I have a Shimano 105 rear derailleur that doesn't seem to be working at all. I have tinkered and tried adjusting but to no avail. The thing will almost not shift at all; sometimes it will work for a small cog to a larger one, but never from a large one to a small one. Like I said, I have played with the adjuster screws and tried tightening the cable, but nothing has worked. Is it just bent?

Well, going from a large cog to a small cog means you are releasing the cable. Sounds like the cable isn't free in the housing.
What have you trird adjusting and tinkering? If you have been turning the limit screws, you may have them set where the thing can't shift.

Whenever I encounter a shifting problem that doesn't respond to normal tuneing the first thing that I do is to check the alignment of the derailleur hanger.

Age/condition of cables and housing?
Explain "tinkered and adjusted".
Depending on the age and condition of your cables and housing they may need replacement. I change mine out about once a year. But I ride 4 to 5 times a week all year 'round, rain or shine. New cables and housing will cost about $20.00. Then follow the Parktool instructions for installation and adjustment.
