10 speed road cassette on 9 speed moutain hub

Looking to upgrade a Surly Cross Check with a 10 speed Shimano road cassette. The hub already on the Cross Check is a Shimano Deore 9 speed hub.

Will it work? Any additional items needed to purchase?

Yes. For rear shifter, shimano MTB and road components at a given speed (8,9,10) are compatible. You may have to mess with spacers behind the cassette and get a narrower 10spd chain.

I assume you're also getting a new 10 speed shifter???

You will need a chain as well. Why are you going 10spd? Just because you have a cassette? I would stick with the 9spd setup. You can buy a road cassette and/or use a MTB cassette for the cross bike if you choose. Do you have a long cage RD? There isn't much advantage going to a 10spd cassette.

I understand the upgrade to the new chain, I also understand the issue of chain line as well.

I'll into account your thoughts on 9spd to 10spd. Maybe going to a 10 just might be just the "new, trendy" thing to do and not actually THAT beneficial in the end.
