You know how not having an umbrella causes rain? And how not bringing a book causes boredom? For me, having only 2 co2 cartridges would cause me to have three flats. I like to have a pump, one or two spare tubes, and a patch kit. I check my patch kit often to count the patches and make sure the glue is still wet!
I have my little frame pump and I never even think about it, but it's always there when I need it. I like to keep 2 spare tubes with me. I got a new saddlebag and it is much smaller than my old one so early this year I only had one tube in there. On my first flat the rest of the ride I was worried about getting a second flat. I squeezed a second tube in there as soon as I got home so I wouldn't have to worry next time. I couldn't fit anything more in there. I got the two tubes wrapped in rubber gloves, tire levers, a couple band-aids, and my multi-tool. If anything happens that can't be covered with those items it would be beyond me to fix it anyway.

I carry a spare tube, patch kit, tire levers and a small multi-tool with hex key and screw driver. As a newbie to clipless peddles I fell and twisted my seat. Glad I could fix that. My chain also broke but I had to call for a ride home for that.
The one other item I have not seen mentioned is a rag to clean your hands with after doing the work.
All this fits nicely in my bag under my seat.

I have my cell phone, some cash, Lezyne Pressure Drive CFH Mini-pump with CO2 capability and one cartridge, multi-tool, zip ties, patch kit, and tire levers. I still need to add a spare tube, knife, some basic sterile 4x4 pads, tape, and some antiseptic ointment. Also, since I'm the rolling pack mule for our family outings, I generally have a few small towels, extra water, and snacks/energy bars, my keys, and all other loose objects that the kids always seem to forget to leave at home or in the car.

I am only set up to deal with flats on my bikes. Anything other than that I am either walking or calling for assistance.
On my road bike & mtb bike I keep the following in a seat bag.
1 spare tube,
1 pack of stick on patches,
1 pliers / multitool,
1 hex wrench set,
few zip ties,
5 dollar bill,
attached to the bike is a mini floor pump.

I also have gloves (I should have listed them in the emergency kit). But in case I forget to restock or I rip one, my hand may get dirty. Sometimes my lock gets on my chain and rubs off some grease, or brake dust. So it doesn't hurt to have some as well. I can also wipe the bike down with them. Or clean my hands before a snack.

I keep a Rema patch kit in my tool pack in case of a really bad day. I also have a few pieces of Gorilla tape on the Rema box for use as a tire patch/boot if needed. I actually used Gorilla tape as a tube patch once; held air just fine for a couple days!
On the subject of Rema kits, I only carry a new, unopened tube of the "glue".
I recently added a pair of tweezers. I had a flat awhile back and the culprit was difficult to get out of the tire.

Aside from flat repair and multi-tool, I carry a chain tool and a few links, spoke wrench, some plastic zip ties, and another little folding tool with pliers and a blade.

Zip ties are a good idea I need to add to my kit. They can fix many things in a pinch. Thanks for the idea Mr. Hamilton.

Frame pump on the frame. Spare tube, patch kit, tire levers, multi-tool in the seat bag. Comes with me on every ride.

Lost a bolt for your rack, zip tie. Pannier clip breaks, zip tie. A few clever zip ties and careful riding may replace a skewer in a real bind (probably will never try this one, but in theory (my theory) it might work).
Citizen arrest bad drivers. Make shift engagement ring for the cute girl on the fixie.
There are probably other legit uses for a quick fix.

Yes zip ties are great, I try to keep a few handy.
Other stuff I have:
Frame pump - for both valve types
Underseat bag with:
1 tube
2 tire levers
multitool
patch kit
CO2 pump & 1 cylinder
1 bus token (or 3 bucks) - which I'm sure glad I had when I blew a tire in -15C weather
If I'm on the beater then I toss a small wrench in my backpack
Other stuff I have:

For my commuter, I have a Topeak Road Morph pump, a tube, a patch kit and some tire levers. Also have a cheap Bell multitool with hex keys, a phillips head screwdriver and a couple of nut wrenches. Since I'm commuting to work I always have a wallet in there too, as well as my cell phone. I keep it all in my backpack. I also carry a u-lock in there.
My commute largely mirrors a bus route that will drop me, literally, a block away from work so if I ever got in a real bind I could do that.
There's also a couple of bike shops not far off my usual route if I needed it, including the shop I bought my commuter from

I couldn't come up with more. There are probably more things that when they come up they will work. I guess the same reason I carry a length of rope. Who knows?
My knife came in handy yesterday. Some fellow couldn't get his coffee beans open in the store to grind them. Bam, here is a knife.

I have a handlebar bag on all my bikes that I got from walmart which cost me about 10 dollars...
it holds:
- tire levers
- multitool
- 2 tubes and patch kit
- a spoke wrench
- chain tool
I haven't needed the last two but I don't want to lose them and they are only in a bag for the commuter.
still have room for my wallet, phone, keys, but I keep them in my backpack, which goes in my "trunk"...

It's kind of like baling wire - you never know but when you need it, it's handy.
Let's say one bolt rattled out of your bottle cage. Maybe mounting bolt broke on your rack. Something happened to the rubber clip on your headlight. Rear shifter problem, zip-tie the cable. Anything you want to fasten up for any reason.

I carry gloves,but I also carry a 1oz squeeze bottle(you can get them from REI) filled with GoJo hand cleaner wrapped with paper towels. I've run into a couple folks who had issues getting their chains back on,plus it's good for getting grease off your clothes and other stuff.

Small pump, tube, levers, patch kit, small adjustable wrench for axle nuts, 5mm Allen wrench for my "DIY anti-theft QR tab", bungee cord, all in my trunk bag. Wallet with ID, some money, Swiss Army knife in my pocket, cell phone on my belt.

Baby wipes are also a wonderful thing. I don't carry them, but I'm considering it. Also, a small length of duct tape or Gorilla Tape™ wrapped around a little stick.

Wrap it around a pen or sharpie. That way you something to write anything down and you can write on the tape. ie license plate, grocery list, orchestral score, name of girl on fixie you proposed to...
