Runtastic does, I do it all the time. With a free account, you can only have 5 custom routes in your profile at a time, but that's fine for most people.

On your bike, you can use any of the variants of the program, they are all the same with different colored skins... the basic Runtastic is Blue, Runtastic Roadbike is Red, Runtastic Mountain Bike is Brown. But from the "activity" list you can tell it you are doing a cycling workout, and all of them have the same options for the various workout types. Other than color of the interface I can't see any difference between the 3 versions.

I tried Runtastic, MapMyRide, Strava, and RideWithGPS; and Runtastic was the winner for me due to it's better map version (it has the OSM Cycling layer available onboard), and somewhat more preferable interface and stats overall.
