does it make a difference over a 12/25 durace 10 speed casette for climbing?

27 is an easier gear than 25. Whether it's enough difference to make you happy is another question. Personally, if I was looking for an easier climbing gear, I'd be looking for a bigger change than that.

A difference, but not much difference. Think about how much easier your current 25 is to the 23 - the 27 will be easier again by about the same. 21-23-25 gets replaced by 21-24-27.

What crankset are you currently running? If it's a double and you need more climbing gears, you'll probably be better off looking at a compact crankset.

I like the campy 13/29. I find it made a difference for me.
