After much experimentation over the winter, I have determined that 7 o'clock is the best valve positioning when adding air from a floor pump. There is the least amount of leakage when fitting the valve.

I use a Lezyne Digital Floor Drive pump with the Pro presta valve head. This setup has no leakage whatsoever, regardless of where the valve is. It also has a bleed valve the head, which makes it easy to let out excess air if I over-inflate the tire. I decided that since I use a pump almost every day, that I should get a good one.

7 to 8 or so seem to work best in the event I have sealant in the tubes

Seems to me that worrying over the best position for the valve on a bike tire is indicative of needing to get a life.

I like to think of myself as a pretty astute thinker. The number of quirky situations I’ve pondered is in the tens of thousands. I can honestly say I’ve never thought about valve positions.

1 o'clock front, 11 o'clock rear. This way they're clear of the fork and stays, respectively, while also nicely positioned for easy application of the twist-on chuck I've been meaning to replace for literally years now. I'm also sure to give the finger to anyone who side-eyes me for doing it this way.

Interesting to note that on my bike with fenders, the 4-5 o’clock positions are best.
