I am wondering if wheels should be changed in pairs? If not, which wheel would make the most noticible difference?

If you're looking for aerodynamics, the front wheel is the one to change. If you're looking for weight, I would imagine that the rear would be lighter just because of less, and lighter spokes than the ones you currently have.

Regardless the answer is the same - no they don't have to be changed in pairs but it does seem to make people feel better when things match.

Yep, I also like for bars, stems and seatposts to match - though, none of my bikes are currently matching.
