No. Shift the rear shifter to the smallest cog. Most rear shifters have number 1 as the largest cog not the smallest. You don't have to touch the front shifter

You have to match the shifter to the derailleur. A 10 speed mountain bike shifter will not work with any 10 speed road derailleur. The cassette is not the important part of the puzzle
