The rear wheel has a hex nut on the right side with the shifter chain going into it. I don't see how you loosen nuts on both ends of the wheel and the wheel would drop out so you can change the tube.
Disconnect the cable from the shifter chain. Then you can loosen the nuts on each end of the rear wheel axle.
Replace the tube. reinstall the rear wheel. Reattach the shifter chain to the shifter cable. Adjust for proper shifting.
