I may be naive in thinking that this is a reasonable first ride of significant distance to attempt but Im always up for an adventure any help/advice/input would be appreciated!

How many days riding do you anticipate? Your comment about a weekend puts your ride in the "Crush the Commonwealth" category. Current recordholder in that race completed Pittsburgh to Philly in 36 hours.

I suggest starting in Philadelphia, taking the Schuykill River Trail to Mont Clare, and then picking up Bike Route "S" in Phoenixville. You can follow the S route to Ohiopyle, five or so miles from Fallingwater. BTW, many people think riding a bike to Fallingwater a bad idea, since the roads have no shoulders and there are a lot of coal trucks using them. Be prepared for serious hill climbing once you pass Phoenixville.

An overnight trip beforehand is useful to help you decided what you need and what you don't, what works and what doesn't.
