I have both the Mille S5 and Uno S5 and wear L in both (185cm, 67kg).

The pads are very similar if not the same but the Mille has that double chamois--anti friction layer, which is sewn in a more curved fashion instead of flat on the Uno. Also there is slightly more chamois in the crotch area in the Mille S5.

The Mille has a longer inseam. The Uno has more compression in the legs. Also the strap construction on the Mille is better in terms of ventilation.

Overall, the Mille S5 is better but very marginally, especially if taken into account the price difference.

If I had to buy another pair I would go for the Mille simply because I ride a lot and can justify the cost but basically, the Unos are pretty much competitive to any of the best other manufacturers put out there including Castelli, rh+, etc.
