Took me about 4-6 weeks of riding 5+ days a week. Took about 2 weeks to get the saddle position correct which probably led to more soreness than necessary. I also had a pro-fit after two weeks.

I had soreness in the sit bone area along with saddle sores which I treated by keeping them clean (Witch Hazel or Rubbing Alcohol) and using liberal amounts of Neosporin + w/Pain Relief Along with A&D Ointment.

I did RAGBRAI on my new saddle that I purchased about 6 weeks prior. No problems at all. Even doing the optional Karras Loop for a 103.5 miles on Tuesday. I felt like I could go another 100 miles.

I use a Specialized Romin Comp Gel. The Gel and padding are very minimal in this saddle. Prior to getting the Romin Comp, I used a very overstuffed ARS Saddle by Planet Bike so I went from super padding to almost no padding.

Edit to add that I used a lot of chamois butter during this transition.

I don't use it at all now. I did use it on the first three days of RAGBRAI just because I was still transitioning in my mind. I think it helped on the century ride. I haven't used it since.
