She also likes the idea of STI although I'm familiar with the modern shifters (high priced) - Campy/Sram/Shimano although what would be a good STI that's at a good price/quality ?

Would it be okay to run the Avid BB7's on them as well ?

Also, I was looking into the 650B's due to the tyre selection (Hetre) weight, volume & all-around profile although anyone have any thoughts with 650B & 26 wheels; I love the VO wheels although they aren't disc !

Going to be for commuting & long rides (60-100 miles per ride) with some touring here and there.

Sram doesn't make a triple crank road shifter. Campy is probably too expensive and you'd need a Campy drivetrain...more expensive. Shimano STI works well and is very durable in my experience. I have (or have had) Tiagra, 105 and Ultegra. While there is some finish difference and some weight difference, it's not all that much. Tiagra works well and will probably serve you for quite a while. My current touring bike has a 46/36/20 crank with 105 shifters, if that is any help.

Since you are planning on the BB7 road discs, they should work with STI because that's what they are designed to do.

You can find lots of 26" tires that will do the job. There are more 650's out there than there used to be but they haven't hit the big box stores yet. They may in the future but I wouldn't expect it for sooner than 5 to 10 years. It is something to consider if you are planning on touring.
