I want the bike to fit 700C X 50 tires with fenders - in other words, Schwalbe Big Ben. No bike today offers the capability to mount fat tires under fenders.

The rear stays have to be wide enough to accept them. I'd want horizontal dropouts to take any kind of gearing from fixed gear to IGH.
