Yes, but if the brakes are hydraulic you have to be careful not to operate the brake lever with no disc rotor in place. You can prevent any problems by inserting a spacer into the place where the rotor fits through the calliper, these are used on bikes being shipped to bike shops in their boxes. So why haven't (or have they?) brake manufacturers made some simple lock out lever or mrchanism so you can't squeeze the handle when you remove the wheel? Sort of like brake quick releases on caliper brakes.
