50 psi in a 2” tire shouldn’t shift. If you had a slow leaker flat, you could drop pressure enough to get some tube creep which might cause your problem. Additionally if the valve stem weren’t at 90° to the rim when you pumped up the tire, that can stress the valve stem as well. It’s very common to cut an angled valve stem, even with proper inflation.
