If you're comfortable with "very hilly" 50 mile rides (say 5000 feet of climbing in 50 miles), then a flat 100 mile ride should be easy (though maybe boring). Just make sure you eat and drink enough during the ride. Your rear end may get worn out from all that sitting, but your legs and lungs should be fine.
