I use DeLorme (5.0) which has its pros and cons. I think it's about $50 (more for more regions of the country).

It does elevation profiles (you've probably seen them - they come out green and brown by default), and you can even mouse over the route and see the continuous grade, etc. plus 3D rendering, etc.

That said, it has maybe one of the world's worst user interfaces - it's not at all intuitive, and some of the maps it prints are less than useful. I use it primarily for scoping out a route as the detail is quite good - it pretty much does have everything. But whether it outweighs the tedium and frustration is a question you'll have to answer.
