Not sure what you mean by soft spot.
Are the tire's beads completely seated in the area of the valve stem. Sometimes the tube gets caught between the tire's bead adjacent and the rim adjacent to the valve stem. If this happens you can push the valve into the tire while deflated, this will remove the tube from under the bead(s).
Or maybe you just have a defective tire.
