Here's what I would suggest -- work back from the date of your MS150 -- 2 weeks before that weekend, you should plan on riding maybe 50 miles on Saturday, and 40 or 50 on Sunday, and 3 weeks before that, maybe 45 / 40 or something like that, and work your way back to this week (maybe try to start with 5-10 miles), building a little each week (typically not more than 10-20% increase in total mileage each week). Your long rides on the weekend (or whenever you have the time available) will be the most important rides you do to prepare. I would also try to get in one or two shorter rides during the week if you can. That would give you a couple days to train for your 5K run and make sure you have a day off or at least a "recovery" day where you just get on the bike and ride very easy for a bit. You probably don't need to start with back to back long rides, but I would make sure you get a least 2 or 3 weekends where you do them so you're used to it before the MS150.
