I did this route on a TnT training ride last Spring - although we went up Sierra instead of Calaveras. We ended up closer to 10,000 feet of climbing, but we used a rather inventive route back to Livermore (the coaches mapped out every hill in Pleasanton, as far as I can tell). My legs hurt for several days afterwards - but that was one of the more enjoyable rides we did last year.
Bring your large water bottles - as someone else noted, there's only one place along a 45 mile stretch to refill your water bottles.
