I've ridden the Tufos with their sealant in the rear tire.

The sealant that is supposed to seal is just pouring out. I tried put the puncture up or down or sideways, and the sealant just came out the hole without sealing.

This is contrary to my experiences - I have used the sealant twice on different tires and they are still going strong. I suspect my method of application is different, though. I would never use quantity of sealant enough to be puring through the hole. Tufo warns against using the sealant as a precaution and does not recommend it for punctures bigger than 2mm.
